Guanidine Hydrochloride denatures proteins and inhibits nucleases. Beta-Mercaptoethanol or DTT increase the denaturing activity of guanidine hydrochloride. Strongly hydrophobic proteins, like the membrane protein lipophilin, will not be denatured by guanidine hydrochloride.
